It starts at an intersection with the US 27 mainline in Columbus in Muscogee County, and initially runs northeast into Harris County, concurrent with Georgia State Route 85 (SR 85). The route splits for SR 85 south of Shiloh, on the Harris–Talbot county line, before it cuts across the northwestern corner of Talbot County. It then enters Meriwether County. Heading briefly northwest through Warm Springs, now co-signed with SR 41, the route turns north and heads through Greenville and Luthersville into Coweta County. US 27 Alternate intersects US 29/SR 14 in Moreland, where they begin a short concurrency to the northwest. They cross Interstate 85 (I-85) southeast of Newnan, where the concurrency with US 29/SR 14/SR 41 end. Then, US 27 Alternate/SR 16 head northwest out of Newnan. They run through Whiteburg in Carroll before reaching its northern terminus, an intersection with US 27/SR 1/SR 16/SR 166 in Carrollton.
